Alteraciones en estos complejos se han relacionado con la patogénesis de múltiples enfermedades autoinmunes y autoinflamatorias. En esta revisión sistemática se analizó la evidencia científica disponible sobre el papel patogénico del proteasoma en patologías como artritis reumatoide, lupus eritematoso sistémico, síndrome antifosfolípido, enfermedad de Sjögren, esclerosis múltiple, tiroiditis autoinmune, enfermedad inflamatoria intestinal y síndromes PRAAS, entre otros. Se destaca la participación del inmunoproteasoma en la diferenciación de linfocitos T y en la producción de citocinas proinflamatorias, así como el impacto de mutaciones genéticas en subunidades proteosómicas. Finalmente, se discute el potencial terapéutico de inhibidores del proteasoma, como bortezomib y ONX 0914, que han demostrado eficacia en modelos preclínicos, aunque requieren estudios adicionales para definir su seguridad y aplicabilidad clínica.]]></p></abstract>
<abstract abstract-type="short" xml:lang="en"><p><![CDATA[ABSTRACT The proteasome and its immunoproteasome variant play essential roles in intracellular protein degradation, cell cycle regulation, and antigen presentation. Dysregulation of these complexes has been linked to the pathogenesis of various autoimmune and autoinflammatory diseases. This systematic review analyzed the available evidence regarding the pathogenic role of the proteasome in conditions such as rheumatoid arthritis, systemic lupus erythematosus, antiphospholipid syndrome, Sjögren&#8217;s syndrome, multiple sclerosis, autoimmune thyroiditis, inflammatory bowel disease, and PRAAS syndromes. The immunoproteasome is highlighted as a key regulator of T-cell differentiation and pro-inflammatory cytokine production, while genetic mutations in proteasome subunits have been associated with autoinflammatory syndromes. Finally, the therapeutic potential of proteasome inhibitors such as bortezomib and ONX 0914 is discussed, showing promising preclinical results but requiring further studies to establish their safety and clinical applicability.]]></p></abstract>
