Abstract: The lapacho negro is a forest species of great importance, it is timber and ornamental, so the establishment of a protocol for micropropagation of Handroanthus heptaphyllus could become an alternative for the mass production of this species. The experimentation was carried out in the Biotechnology Laboratory of the Biology Area of the Facultad de Ciencias Agrarias de la Universidad Nacional de Asunción, with the general objective of establishing a protocol for the in vitro micropropagation of Handroanthus heptaphyllus from nodal segments. For the population of variables and units, 1620 explants were planted, each experimental unit consisted of 1 nodal segment. The experimentation had 12 treatments sown in two culture media (Agar/Water and MS), which were based on variations in immersion times of ethanol and sodium hypochlorite for contamination control, for oxidation control the treatment was applied of oxidation prevention 2 g/l of activated carbon, and phytoregulators (AIB and BAP) were added. The mother plants were treated with Copper Oxychloride (OxiCob®) and Dithiocarbamate (Mancozeb®) in a proportion of 3 and 2 g/l respectively. The variables studied were the control of leaf spots in mother plants, contamination of explants, oxidation, survival, sprouting, callus formation and the length of the shoots. Statistical analysis consisted of Kolmogorov-Smirnov normality tests for all variables. 45.8% leaf spot control was achieved, 50% contamination control, oxidation was 100% controlled. 60% of living explants were obtained up to 50 days with leaf differentiation of 1.5 mm of average elongation. Based on these results, it is concluded that the applied treatments affected the variables studied.
