The Ypacaraí Lake is a valuable water resource of Paraguay, it is the largest lake in the country and is known internationally through the music and tourism. The great demographic growth and anthropogenic activi- ties developed in its basin have caused its contamination and environmental degradation in recent years. The objective of this work was to develop and validate a method for the determination of di(2-ethylhexyl) adipate (DEHA) in its surface waters. The liquid-liquid dispersive microextraction technique (DLLME) optimized by means of a multivariate experimental design was used within the analytical methodology. The validation of the analytical method was carried out an evaluation of each of the characteristics of a performance following the guidelines of the Eurachem 2014 guide. The developed, optimized and validated methodology also applies to the determination of DEHA in the waters of Ypacaraí Lake. The limit of detection LOD and limit of quantification LOQ were 0.004 mg.L-1 and 0.014 mg.L-1 respectively. The precision of the method evaluated by repeatability and intermediate precision was 2.37 % and 3.78 % and 87.14 % of recovery at LOQ level and 97.62 % at 0.3 mg.L-1. The results include the presence of this contaminant in them between 0.028 mg.L-1 and 0.603 mg.L-1. The analytical method was successfully developed, optimized and validated and it enabled the correct quantification of the DEHA compound in the surface waters of Ypacaraí Lake.
