Abstract

SANABRIA, Diana; GIMENEZ, Vivian; CARPINELLI, María Mercedes  and  ROLON, Julio. First experience in Paraguay of  reference values determination using the dihydrorhodamine (DHR) assay in the evaluation of the neutrophil respiratory burst in healthy children. Pediatr. (Asunción) [online]. 2016, vol.43, n.1, pp.33-38. ISSN 1683-9803.  https://doi.org/10.18004/ped.2016.abril.33-38.

Introduction: Chronic Granulomatous Disease (CGD) is a primary immunodeficiency characterized by an impaired neutrophil functionality, essential in humans for the defense against infections. The nitroblue tetrazolium test (NBT) is useful for diagnosis, standing out among currently applied flow cytometry tecniques like the dihydrorhodamine test (DHR). Objective:: To establish the range of reference values for the DHR assay in the evaluation of the neutrophil respiratory burst in healthy children. Materials and Methods: A descriptive cross-sectional observational study with an analytical component. Fifty four apparently healthy children who were under 16 years of age, that attended the pediatric consultation for a general health state check-up or pre-surgical reasons at the pediatric offices of the  Barrio Obrero General Hospital, were included. Neutrophil functionality was evaluated by NBT and DHR techniques. Results: NBT reduction values above 85% were observed in all children. The reference range established for neutrophil stimulation index (SI) by the DHR assay was 22.5 to 60.4 with a mean of 35.9 ± 10.1. Statistically significant differences were not found between the means of SI of age ranges (p = 0.336) nor sex (p = 0.174). Conclusion: The range of reference values established for the DHR assay in the evaluation of the neutrophil respiratory burst in the studied healthy children population differs from others found in literature.  This shows the importance of establishing values that consider the characteristics of the study population and the methodology of each laboratory.
